The substrate is supplied as two components; color solution (A) and stabilizer concentrate (B). The product is designed to be reconstituted with water to complete the manufacturing process and to produce a ready to use solution (TMB). The reconstituted product is a colorless liquid or very light yellow in appearance. When TMB is reacted with peroxidase, a reaction ensues producing a blue colored liquid product (oxidized state). A reagent (blue stop) tailored to TMB to stop the reaction at this “blue stage”. The reaction also can be terminated more typically using an appropriate reagent (an acid) or proprietary Yellow Stop specifically designed for TMB Substrates. By stopping the reaction in this manner, a second oxidized state is produced resulting in a soluble yellow reaction product. Depending on the stop reagent utilized, the colored product is stable for at least two hours. TMB is not recommended for membrane or immunohistochemistry applications where a colored precipitation reaction is desired. Product Use: The concentrated substrate is supplied as two components and designed to be reconstituted with water to complete the manufacturing process to produce a ready to use one component TMB solution. Equilibrate the product to RT (25°C) before use. Occasionally, due to the high concentration of solutes in component B, crystals may form during storage. Reconstitute component B with pure ASTM type 1 RODI water (9X the original component B volume). Gently agitate the resulting solution on occasion and equilibrate for at least 60 minutes or until all particulates are dissolved. Next, add Component A color solution to the reconstituted B stabilizer solution. Gently agitate the resulting solution on occasion and equilibrate for 60 minutes. Equilibrate this final solution overnight at room temperature and gently agitate a few times the next day. The final solution is now a completed product (TMBST). For immunoassay microwell applications, 100ul of substrate solution should be added to each well. A liquid soluble blue reaction develops which can be read at ~370nm or between 620nm and 650nm. Optimally, absorbance values should be monitored before they exceed 2.0OD units. In endpoint assays and for best results, the proprietary Yellow Stop reagent should be used. The substrate reaction also can be stopped using equal volumes of 1N HCL or 0.6N Sulfuric Acid. Addition of Yellow Stop or acid turns the blue color to yellow at end point. With Yellow Stop, absorbance values increase approximately three fold after addition. Hence, sample OD values should be monitored and the substrate reaction terminated when values reach ~0.7OD units. For Yellow Stop or an acid Yellow Stop, a liquid soluble yellow reaction develops which is read in the 450nm range. Dilution of the substrate is not recommended and will alter its performance characteristics. To reduce reaction kinetics, it is recommended that analytes, antibodies or conjugates used in the assay be diluted. TMB is light sensitive and should be protected from direct sunlight or UV sources. Storage and Stability: Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
