Description TIMP-1 is the first described and best characterized of the four known mammalian TIMPs. The TIMPs are so named because they are slow, tight binding endogenous inhibitors of the Matrix Metalloproteinases (MMPs). The four TIMPs have different inhibition constants for the different MMPs studied. TIMP-2 has a greater efficiency against MMP-2, and TIMP-1 works better against MMP-9, MMP-1 and MMP-3. The function of the ADAMs (A Disintigrin And Metalloproteinase) proteinases is getting more attention, and it has been shown that TIMP-3 is an efficient "sheddase" inhibitor, inhibiting ADAM-17 (TACE) at the low nanomolar levels seen with MMP inhibition. The other ADAMs proteinases have not yet been assayed for TIMP inhibition, but TIMP-1 seems to be much less active on ADAM-17 than is TIMP-3. The TIMP-1 is largely the glycosylated form, about 29kD in size on reduced Western Blots. T5580-06B is recombinant mouse tissue inhibitor of metalloproteinases-1 (rmTIMP-1) with a histidine tag. Applications: Suitable for use in Enzymatic Assays, ELISA, Western Blot and Substrate Gel Analysis. Other applications not tested. Recommended Dilution: Western Blot: Resuspend the lyophilized powder in 250ul SDS-PAGE sample buffer. Load 10ul per lane. Standard can be detected as a 32kD protein under reducing conditions and as a 30kD protein under non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
