Description GM6001 MMP Inhibitor (Ilomastat) is a potent inhibitor of collagenase. Broad spectrum inhibitor of matrix metalloproteases (MMPs) with Ki factor in nM range. Widely used to inhibit MMPs in vitro experiments and in animal models. Nomenclature: N-[(2R)-2-(hydroxamidocarbonylmethyl)-4-methylpentanoyl]-L-tryptophan methylamide Reported Ki values are as follows (see Galardy Ref): Human MMP-1 (Fibroblast collagenase): 0.4nM Human MMP-3 (Stromelysin): 27nM Human MMP-2 (72kD gelatinase): 0.5nM Human MMP-8 (Neutrophil collagenase):0.1nM Human MMP-9 (92kD gelatinase): 0.2nM Applications: Suitable for use in Cell Culture Recommended Dilutions: Cell Culture: 10-25uM It is recommended that the concentration of GM6001 in aqueous working solutions is determined upon dilution and confirmed prior to each use. Relative concentrations of GM6001 in aqueous solutions may be determined by measuring the absorbance of the solution (tyrosine) at 280nm. Reconstitution: Reconstitute in DMSO (at least 25mM), aliquot and store at -20°C. Other organic solvents (ethanol) have also been used. Further dilutions can be made with dH2O or with low salt buffers. G8237 will precipitate from aqueous solutions exceeding 100uM. Aqueous solution will hydrolyze or decompse at a rate of 1-2% per week. Sonication of the mixture will enhance dissolution. Quality Control: H-NMR Spectrum: Conforms Mass Spectrum: 389 [M+1] Thin Layer Chromatography: One spot (ethyl acetate:Methanol = 6:1) Storage and Stability: Powder is stable at RT for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
