Description DNA Marker Ladder Mix, 21 Fragments (100-10000bp), Ready to Use, BioAssay™ The ladder is composed of 21 chromatographically purified DNA fragments (in base pairs): 10000, 8000, 6000, 5000, 4000, 3500, 3000, 2500, 2000, 1500, 1200, 1000, 900, 800, 700, 600, 500, 400, 300, 200, 100. The bright bands at 3000, 1000, and 500 bp serve as reference bands. Band Concentration: 0.5ug of DNA Ladder Mix contains: a) 60ng (12.0%) for each of the 500, 1000 and 3,000bp reference fragments. b) 20ng (4.0%) for 100-400 bp bands c) 17ng (3.4%) for 600-900 bp bands d) 16ng (3.2%) for 1200-2600 bp bands e) 18ng (3.6%) for 3500-10,000 bp bands. Storage and Loading Buffer: Supplied as a liquid in 10mM Tris-HCl, pH 7.6, 10mM EDTA, 0.005% bromophenol blue, 0.005% xylene cyanol FF, 10% glycerol. 6X Loading Dye Solution L3350: (Included) Supplied as a liquid in 10mM Tris-HCl, pH 7.6, 0.03% bromophenol blue, 0.03% xylene cyanol FF, 60mM EDTA, 60% glycerol.. Quality Control: Well-defined bands are formed during agarose gel electrophoresis. The absence of nucleases is confirmed by a direct nuclease activity assay. Recommendations for Use: 1. Vortex gently just prior to use. 2. Do not heat before loading. 3. Dilute DNA sample with L3350, 6X DNA Loading Dye; mix 1 volume of dye with 5 volumes of DNA sample. 4. Apply 1ul (0.1ug) of DNA Ladder (D3930-14A) per 1mm of agarose gel lane width. 5. For quantitation, adjust the concentration of the sample to match the amount of DNA in the nearest band of the ladder. 6. Visualize DNA bands by staining with ethidium bromide. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
